2021年と2022年に盛岡市内の数地点でキタキチョウを採集し、ボルバキア感染状況を調査した。さらに、昨年度報告した、判別用プライマー(竹内・佐原, 2021)を用いて、感染個体と非感染個体におけるミトコンドリアの由来の特定を試みた。

rDNAクラスターの染色体上の位置は、rDNAを含むプローブを用いて染色体標本にFISH(fluorescence in situ hybridization)を行うことで視覚的に確認できる。特に18S rDNAは配列保存性が高いため、コドリンガ(Cydia pomonella)のgDNAを用いたPCRにより18S rDNAを含む1,650 bpの配列を増幅し、増幅産物にラベルしたプローブが広く利用されている(FUKOVÁ et al. 2005)。本研究では、18S rDNAを含むタンデムrDNA配列がクローンされたと考えられるオオモンシロチョウBACクローンを発見したので、他種へのrDNAクラスター検出に用いることが可能かどうか検証した。

OKUMURA et al.(2019)が構築したコウモリガBACライブラリーの総インサートサイズは約2 Gbと推定される。ゲノムリダンダンシーを考慮すると、小林(未発表)の選抜した、カイコ単一遺伝子オルソログを含むコウモリガBACクローンが69に留まったのは、ライブラリーのゲノムカバー率の低さが影響した可能性も否めない。染色体の対応関係を網羅的に比較するためには、1染色体あたり4以上のマーカーを必要とする。小林(未発表)では、カイコ単一オルソログ含むBACを複数獲得できていない染色体が存在し、研究完遂にはさらなるBAC選抜が必要と考えられた。そこで、本研究では新規のカイコオルソログ含有コウモリガBACを効率的に選抜するために新規コウモリガBACライブラリーの構築を行った。

SxlIn3-homoの有核精子に異常がないことは、3倍体雄との重複交尾により確認された。そのため、この方法による系統維持は可能である。しかし、系統維持の度毎に3倍体を誘起することはスペースや労力的に負担が大きかった。そこで、黒縞遺伝子座(pS)をホモにもつre9系統をヘルパーとして利用する方法が開発された(SAKAI et al. 2020)。この研究では、卵色を用いたre9の交尾時間の最適化とSxlIn3-homo雌への確認実験を行った。最適な重複交尾の結果、SxlIn3-homo個体と考えられる次世代の形蚕ホモ(+p/+p)個体割合は0~31.5%、最大受精卵数は112個であった。この研究での実験区数は6と小さく、形蚕個体の受精については確認されていなかった。そこで本研究では、これら次世代の単交尾における受精率がゼロであるSxlIn3-homoだと証明するとともに、その次世代へも系統維持できることを示す目的で実験を行った。

非感染キタキチョウの由来の特定は、ミトコンドリア配列を確認すればよい(NARITA et al. 2006; MIYATA et al. 2017; 2020)。しかし、大量の個体を配列特定するにはコストがかかる。そこで本研究では、ミトコンドリア遺伝子配列がキタキチョウ型かミナミキチョウ型かを簡便に識別できるPCR法の確立に取り組んだ。先行研究(NARITA et al. 2006)で公表されているキタキチョウおよびミナミキチョウのND5配列を参考にプライマーを設計し、キタキチョウ型とミナミキチョウ型の簡易識別に成功した。

S. tienmushanensis(St)のゲノムデータについてヒゲナガカワトビケラ染色体上へのオルソログマッピングを通じた染色体レベルのゲノムアセンブルを目指した。本研究では、Stコンティグデータとヒゲナガカワトビケラの配列情報を対応付けるために、ヒゲナガカワトビケラ蛹個体から抽出した全RNAを用いてRNAseqを行った。さらに、獲得した配列情報を用いてカイコ遺伝子と1対1対応のオルソログもしくはStコンティグデータに対応する配列を持つヒゲナガカワトビケラのBACセレクションを行った。

Sex is determined by diverse mechanisms and master sexdetermination
genes are highly divergent, even among closely
related species. Therefore, it is possible that homologs of master
sex-determination genes might have alternative functions in
different species. Herein, we focused on Sex-lethal (Sxl), which is
the master sex-determination gene in Drosophila melanogaster
and is necessary for female germline development. It has been
widely shown that the sex-determination function of Sxl in Drosophilidae
species is not conserved in other insects of different
orders. We investigated the function of Sxl in the lepidopteran
insect Bombyx mori. In lepidopteran insects (moths and butterflies), spermatogenesis results in two different types of sperm: nucleated fertile eupyrene sperm and anucleate nonfertile parasperm, also known as apyrene sperm. Genetic analyses using Sxl mutants revealed that the gene is indispensable for proper morphogenesis of apyrene sperm. Similarly, our analyses using Sxl -

We constructed the first trichopteran bacterial artificial chromosome (BAC) library in Stenopsyche marmorata. The BAC library consists of 32,256 clones with an average insert size of 65.38 kb. We also made a cDNA library from an S. marmorata adult male and determined 598 unique sequences. Using the obtained sequences we designed putative single tagged site (STS) primers and selected 81 BACs containing orthologs of Bombyx mori genes by three-step PCR selection method. These BACs will be used for gene-based comparative mapping by fluorescence in situ hybridization (FISH) to identify regions of conserved synteny between the trichopteran and lepidopteran genomes. -

Antheraea yamamai is a wild silkmoth that spins yellow-green silk to produce cocoons. Cocoons that are less yellowish in color, referred to as emerald green (EG), have been found in A. yamamai mass-rearing for cocoon production. Here, we examined EG inheritance by crossing individuals that produce EG cocoons with those that produce normal-colored (normal green: NG) cocoons. F1 progenies from the reciprocal cross between EG and NG individuals mostly produced NG cocoons. In the backcross, color
segregation mostly indicated that the EG mutation is controlled by a single recessive gene locus. Single-pair selection of cocoon weight for 4 years revealed that there is a heterosis effect on cocoon weight in the hybrid
progenies. -

We have developed a fast and efficient PCR-based technique to sex all developmental stages of the ladybird beetle Harmonia axyridis. Previously, we established a male-specific fluorescent expressing strain of H. axyridis with the piggyBac vector. FISH analyses revealed that the transgene inserted into the H. axyridis genome in the male Y-chromosome. Because only males expressed the fluorescent signal, we were able to differentiate between the sexes of this transgenic strain even in newly hatched larvae. We took advantage of this insertion and designed an inverse PCR reaction and genome walking surrounding the inserted region to identify 3,781 bp of the male Y-chromosome genomic sequence. From this male specific region, we designed a PCR-based protocol to identify sexes from all H. axyridis developmental stages.

ガとチョウ類のFISHを通じた比較ゲノム解析を進めるため、重要な農業害虫であるヨトウガのフォスミドライブラリーを作製し、EST配列の決定からカイコ単一遺伝子のオルソログを含むヨトウガフォスミドを120個特定した。この研究ではcDNAの行列プールを用いることでEST配列決定の効率化を実現した。一部のフォスミドを用いたFISH解析よりカイコとのシンテニーが示された。

This is the first report of tandemly arrayed sex pheromone receptor genes in Lepidoptera. The localization of an OR gene cluster on the Z chromosome agrees with previous findings for a Z-linked locus responsible for O. nubilalis male behavioral response to sex pheromone. The 181-bp irect repeats might enhance gene duplications by unequal crossovers. An autosomal locus responsible for male esponse to sex pheromone in Heliothis virescens and H.subflexa was recently reported to contain at least four OR genes. Taken together, these findings support the hypothesis that eneration of additional copies of OR genes can increase the potential for male moths to acquire altered specificity for pheromone components, and accordingly, facilitate differentiation of sex pheromones.

We performed gene-based comparative FISH mapping between a wild silkmoth, Samia cynthia ssp. with a low number of chromosomes (2n=25-28) and the model species, Bombyx mori (2n=56), in order to identify the genomic components that make up the chromosomes in a low-number karyotype. Mapping
of 64 fosmid probes containing orthologs of B. mori genes revealed that the homologues of either two or four B. mori chromosomes constitute the S. c. ricini (Vietnam opulation, 2n=27/28, Z0/ZZ) autosomes. Where tested, even the gene order was conserved between S. c. ricini and B. mori. This was also true for the originally autosomal parts of the neo-sex chromosomes in S. c. walkeri (Sapporo population, 2n=26/26, neo-Wneo-Z/neo-Zneo-Z) and S. cynthia subsp. indet. (Nagano population, 2n=25/26, neo-WZ1Z2/Z1Z1Z2Z2). The results are evidence for an internal stability of lepidopteran chromosomes even when all autosomes had undergone fusion processes to form a low-number karyotype. -

Lepidoptera, butterflies and moths, is the second largest animal order and includes numerous agricultural pests. To facilitate comparative genomics in Lepidoptera, we isolated BAC clones containing conserved and putative single-copy genes from libraries of three pests, Heliothis virescens, Ostrinia nubilalis, and Plutella xylostella, harboring the haploid chromosome number, n = 31, which are not closely related with each other or with the silkworm, Bombyx mori, (n = 28), the sequenced model lepidopteran.
A total of 108–184 clones representing 101–182 conserved genes were isolated for each species. For 79 genes, clones were isolated from more than two species, which will be useful as common markers for analysis using fluorescence in situ hybridization (FISH), as well as for comparison of genome sequence among multiple species. -

Individuals of the social spider mite Stigmaeopsis longus live communally in narrow, humid nests made from silk threads and display nest sanitation behaviour through the coordinated deposition of faeces. We used artificial dust to experimentally determine that females of this species use silk threads to perform regular cleaning of the nest space and eggs. We first learned that silk-weaving behaviour is not a by-product of nest building (nest reinforcement); rather, it is actively performed as a function of cleaning the living space and eggs. Furthermore, we determined the effectiveness of the attending females by artificially manipulating their natural habitat, which is clearly connected to the cleaning behaviour by parental females. As such, we have uncovered an extraordinary new role of silk threads as devices
for cleaning the nest space and/or eggs. -

B. mori chromosome 11 corresponds partly to another
autosome (A2) and partly to a chromosome carrying
nucleolar organizer region (NOR) of this subspecies. The
NOR chromosome of S. c. ricini is also partly homologous to
B. mori chromosome 24. -

The baculovirus Autographa californica multiple nucleopolyhedrovirus (AcMNPV) is used as a safer viral vector in mammalian cells with potential applications in gene therapy. However, the mechanism for the insusceptibility of mammalian cells to proliferative infection by entomopathogenic viruses is not well understood. Here, we studied the significance of epigenetic modifications such as histone acetylation, histone methylation and HP1 accumulation for AcMNPV gene expression in mammalian BHK cells. Real-time PCR and chromatin immunoprecipitation with sodium butyrate revealed an important relationship between viral gene expression and histone acetylation, with implications for a mechanism of suppression of AcMNPV gene expression in BHK cells.

We examined chromosomal distribution of major ribosomal DNAs (rDNAs), clustered in the nucleolar organizer regions (NORs), in 18 species of moths and butterflies using fluorescence in situ hybridization with a codling moth (Cydia pomonella) 18S rDNA probe. Most species showed one or two rDNA clusters in their haploid karyotype but exceptions with 4–11 clusters also occurred. Our results
in a compilation with previous data revealed dynamic evolution of rDNA distribution in Lepidoptera except Noctuoidea, which showed a highly uniform rDNA pattern. In
karyotypes with one NOR, interstitial location of rDNA
prevailed, whereas two-NOR karyotypes showed mostly
terminally located rDNA clusters. A possible origin of the
single interstitial NOR by fusion between two NOR-hromosomes
with terminal rDNA clusters lacks support in available data. In some species, spreading of rDNA to new,
mostly terminal chromosome regions was found. -

Feasible methods for sperm cryopreservation and subsequent artificial insemination are urgently needed to maintain
silkworm genetic resources with less laborious work and space cost. We have discovered that loss of the fertilizing
ability of cryopreserved diploid (p50) semen was restored by the addition of either fresh or frozen apyrene sperm
from triploid donors. To decrease usage of the amount of stored diploid semen, we monitored the effective ratio of
cryopreserved p50 semen and apyrene sperm from triploid males to restore fecundity. Restoration of the fertilizing
ability of cryopreserved p50 semen was kept by roughly one fourth decrease of the amount by increasing the apyrene
sperm ratio. Because semen collection from diploid males requires skillful and careful techniques, decreasing the
amount of cryopreserved semen may practically save laborious work. -

Similar nest-weaving habits and sociality characterize all the species belonging to the recently reinstated spider mite genus Stigmaeopsis. Molecular phylogenetic analysis using 28S rDNA shows that Stigmaeopsis forms a distinct clade and that there is another unrecognized clade involving
Schizotetranychus species with comparable nest-weaving habits distributing in tropical regions. Furthermore,

Multicolor fluorescence in-situ hybridization (FISH) and subsequent reprobing of chromosome preparations increase the number of chromosomes and/or anchor loci on the chromosomes simultaneously identified. Reprobing techniques have been widely applied to chromosomes of vertebrates and plants. We have developed a novel reprobing protocol that utilizes multicolor FISH and bacterial artificial chromosome (BAC) probes to examine chromosome preparations in a model lepidopteran species, the silkworm, Bombyx mori. With standard two-color BAC-FISH, routinely used to map genes on B. mori chromosomes, we could localize only two probes on one preparation,
whereas our new protocol combining five-color BAC-FISH and preparation reprobing enabled us to simultaneously map 10 probes, as demonstrated with the Bombyx Z chromosome. The improved BAC-FISH technique will facilitate karyotyping and synteny analysis in Lepidoptera. -

A novel cry gene, cry8Db, highly toxic to scarab beetles such as the Japanese beetle, Popillia japonica Newman, was cloned from an isolate of Bacillus thuringiensis(Bt), BBT2-5. The cry8Db gene has 3525bp nucleotides and codes for a protein of 1174 amino acid residues. The protein, Cry8Db, has typical Bt characteristics such as the 8-block, conserved sequences and the three-domain 3D toxin structure as defined with Cry3Aa.The Cry8Db protein was assayed along with the other scarab active Cry8Da and Cry8Ca against the Japanese beetle. While Cry8Da and Cry8Db had toxicity against both adults and larvae of the Japanese beetle, Cry8Ca was toxic to only larvae. Cry8Ca showed no toxicity against the adult beetle up to 30 microg per 1 cm(2) of leaf discs on which the protein was applied.

The successful assignment of the existing genetic linkage groups (LGs) to individual chromosomes and the second-generation linkage map obtained by mapping a large number of bacterial artificial chromosome (BAC) contigs in the silkworm, Bombyx mori, together with public nucleotide sequence databases, offer a powerful tool for the study of synteny between karyotypes of B. mori and other Lepidopteran species. Conserved synteny of genes between particular chromosomes can be identified by comparatively mapping orthologous genes of the corresponding linkage groups with the help of BAC-FISH (fluorescent in situ hybridization). To achieve higher-throughput comparative mapping using BAC-FISH in Lepidoptera, we developed a protocol for five-color BAC-FISH, which allowed us to map simultaneously 6 different BAC probes to chromosome 15 in B. mori. All 6 M. sexta BAC clones hybridized to a single M. sexta bivalent in pachytene spermatocytes.

We detected a putative gamma-tubulin gene in silico and detected BACs containing the gene from a Bombyx mori BAC library. BAC-FISH mapping revealed that the gene is located on chromosome 5. To observe the distribution of gamma-tubulin, we employed antibodies against mammalian gamma-tubulin peptides. Western blot analysis disclosed a band very similar in size to gamma-tubulin protein in other species (approximately 48 kDa). In mitotic metaphase of B. mori spermatogonial cells, gamma-tubulin is exclusively localized in the spindle poles, where the centrosomes occur. We applied the same system to the grasshopper Chortophaga viridifasciata, as a representative of insect orders in which the gamma-tubulin distribution had not previously been studied. Gamma-tubulin was also found in the spindle poles during metaphase of spermatogonial cells in the grasshopper.

A computer-assisted analysis identified tentative target sequences for regulatory proteins including ecdysone-inducible factors such as BmFTZ-F1 and Broad-Complex Z4 (BR-C Z4) in the ie1 promoter of BmNPV. A transient expression experiment using BmN cells and a series of truncated ie1 promoter constructs demonstrated that the activity of the ie1 promoter responded to alpha-ecdysone and 20-hydroxyecdysone, which required a tridecameric nucleotide stretch (ie1EcRE, 5'-GTGTTATCGACCT-3') homologous to the ecdysone response element reported for Drosophila (DmEcRE). RT-PCR demonstrated the expression of BmEcR and BmUSP, which are required as ecdysone-specific activators for EcRE-mediated activation, in BmN cells. Furthermore, the ie1 EcRE-mediated response was confirmed by using a recombinant BmNPV possessing a luciferase gene under the control of the ie1 promoter with or without ie1 EcRE. This is the first report of an ecdysone response element in a baculoviral gene promoter.

We newly isolated seven W chromosome derived bacterial artificial chromosome (W-BAC) clones from Bombyx mori BAC libraries by means of six different STS primer sets, which amplify each different female specific DNA fragment. Two color fluorescence in situ hybridization (FISH) with 19L6H W-BAC probe as a standard W-chromosome painter disclosed that six out of seven new W-BAC probes strongly highlighted the whole W chromosome in B. mori. Besides the W, hybridization signals occurred in telomeric and/or subtelomeric regions of the autosomes. The remaining W-BAC, 20H9D, provided discontinuous signals, which appeared in both terminal and central compartments of the W chromosome. A representative of W-chromosome painting BACs, 19L6H, and 20H9D W-BAC were hybridized to chromosome spreads of Bombyx mandarina females. The 19L6H W-BAC bound to the whole B. mandarina W chromosome as in B. mori.

We newly identified the W chromosome in eight lepidopteran species and confirmed the absence of W chromosome in Caligula japonica females with the Z0 sex chromosome constitution by means of genomic in situ hybridization (GISH). In pachytene oocytes of species with the WZ/ZZ sex chromosome system, female-derived genomic probes of the respective species highlighted the whole W chromosome thread in the WZ bivalent and also stained specific autosomal region(s) with telomeric and/or subtelomeric signals. On the basis of GISH results obtained in this study and earlier published, we classified karyotypes of lepidopteran species into three types.

Two novel crystal protein genes, cry30Ba and cry44Aa, were cloned from Bacillus thuringiensis subsp. entomocidus INA288 and expressed in an acrystalliferous strain. Cry44Aa crystals were highly toxic to second-instar Culex pipiens pallens (50% mortality concentration [LC50] = 6 ng/ml) and Aedes aegypti (LC50 = 12 ng/ml); however, Cry30Ba crystals were not toxic.

Female moths of Bombyx mori were artificially inseminated with cryopreserved semen. The fertility of inseminated females varied from 0% to 76.9% depending on the strain. Addition of fresh semen from triploid males, which are infertile but whose semen includes intact apyrene sperm, greatly improved fecundity of cryopreserved semen from normal males. Frozen apyrene sperm from the triploid donors also improved the fecundity of females, inseminated with cryopreserved normal semen, but less than fresh semen from triploid males. Fertilization success in B. mori requires the presence of both, intact eupyrene and apyrene sperm. Our results show that eupyrene sperm tolerate the cryopreservation process better than apyrene sperm. Hence, we recommend to add apyrene sperm from the triploid donors as helper sperm routinely to cryopreserved semen in artificial insemination. This may advance the application of cryopreservation as a routine technique to maintain silkworm resources.

A second-generation linkage map was constructed for the silkworm, Bombyx mori, focusing on mapping Bombyx sequences appearing in public nucleotide databases and bacterial artificial chromosome (BAC) contigs. A total of 874 BAC contigs containing 5067 clones (22% of the library) were constructed by PCR-based screening with sequence-tagged sites (STSs) derived from whole-genome shotgun (WGS) sequences. A total of 523 BAC contigs, including 342 independent genes registered in public databases and 85 expressed sequence tags (ESTs), were placed onto the linkage map. We found significant synteny and conserved gene order between B. mori and a nymphalid butterfly, Heliconius melpomene, in four linkage groups (LGs), strongly suggesting that using B. mori as a reference for comparative genomics in Lepidotera is highly feasible.

We cloned Bm-Sxl, an orthologue of the Drosophila melanogaster Sex-lethal (Sxl) gene from embryos of Bombyx mori. The full-length cDNAs were of 2 sizes, 1528 and 1339 bp, and were named Bm-Sxl-L and Bm-Sxl-S, respectively. Bm-Sxl-L consists of 8 exons and spans more than 20 kb of genomic DNA. The open reading frame (ORF) codes for a protein 336 amino acids in length. Bm-Sxl-S is a splice variant that lacks the second exon. This creates a new translation start 138 nucleotides downstream and an ORF that codes for 46 amino acids fewer at the N-terminus. Linkage analysis using an F2 panel mapped Bm-Sxl to linkage group 16 at 69.8 cM. We isolated 2 BACs that include the Bm-Sxl gene. With BAC-FISH we located Bm-Sxl cytogenetically on the chromosome corresponding to linkage group 16 (LG16) at position >68.8 cM.

SXL gene has acquired a pivotal role in the sex-determining pathway of Drosophila, although it does not act as a sex determiner in non-drosophilids. The SXL protein has moderately conserved N- and C-terminal regions and a well-conserved central region including 2 RNA recognition motifs. Our phylogenetic analysis shows that a single orthologue of the Drosophila Sex-lethal (Sxl) gene is present in the genomes of the malaria mosquito Anopheles gambiae, the honeybee Apis mellifera, the silkworm Bombyx mori, and the red flour beetle Tribolium castaneum. The D. melanogaster, D. erecta, and D. pseudoobscura genomes, however, contain 2 paralogous genes, Sxl and CG3056, which are orthologous to the Anopheles, Apis, Bombyx, and Tribolium Sxl. Hence, a duplication in the fly clade generated Sxl and CG3056.

The gene expression of Autographa californica multiple nucleopolyhedrovirus (AcMNPV) was examined in two types of mammalian cells, human HeLa14 and hamster BHK cells. DNA microarray analysis followed by reverse transcription-PCR identified at least 12 viral genes transcribed in both HeLa14 cells and BHK cells inoculated with AcMNPV. 5' rapid amplification of cDNA ends was carried out to examine the transcriptional fidelity of these genes in HeLa14 cells. The transcription of ie-1, ie-0 and gp64 was initiated at a baculovirus early gene motif, CAGT, accompanied by a TATA motif. In addition, the same splicing observed for ie-0 mRNA in Sf9 cells occurred in HeLa14 cells. While the transcription initiation sites for pe38 and p6.9 were not located in the CAGT motif, most of them were in a typical eukaryotic RNA polymerase II promoter structure (a conventional TATA motif and/or an initiator).

In this study, we analysed the W chromosomal regions of the Zebra-W strain (T(W;3)Ze chromosome) and the Black-egg-W strain (T(W;10)+(w-2) chromosome) at the molecular level. Our results strongly indicate that the regions containing the W-Samurai and W-Mikan RAPD markers or the W-Mikan RAPD marker were deleted in the T(W;3)Ze and T(W;10)+(w-2) chromosomes, respectively, due to reciprocal translocation between the W chromosome and the autosome. This deletion apparently does not affect the expression of Fem; therefore, this deleted region of the W chromosome does not contain the putative Fem gene.

We have developed a simple method to resolve the sex chromosome constitution in females of Lepidoptera by using a combination of genomic in situ hybridization (GISH) and fluorescence in situ hybridization with (TTAGG)( n ) telomeric probe (telomere-FISH). In pachytene configurations of sex chromosomes, GISH differentiated W heterochromatin and telomere-FISH detected the chromosome ends. With this method we showed that Antheraea yamamai has a standard system with a fully differentiated W-Z sex chromosome pair. In Orgyia antiqua, we confirmed the presence of neo-W and neo-Z chromosomes, which most probably originated by fusion of the ancestral W and Z with an autosome pair. In contrast to earlier data, Orgyia thyellina females displayed a neo-ZW(1)W(2) sex chromosome constitution. The combination of GISH and telomere-FISH enabled us to acquire not only reliable information about sex chromosome constitution but also an insight into sex chromosome evolution in Lepidoptera.

We previously determined that certain recessive genes decrease female fecundity in a haplo-diploid spider mite, Stigmaeopsis miscanthi (Saito). However, whether the depression was caused by the breakdown of heterosis or the expression of deleterious genes retained in a population could not be determined, because we had started our inbreeding experiment from a mixture of two isolated populations. In order to answer this basic question, inbreeding effects on survival and fecundity were measured for eight small populations occurring far from the two initial populations. There was little depression of immature survival of inbred lineages in all populations. On the other hand, in two inbred lineages, both originating from the smallest populations, female oviposition decreased significantly with the increase of Wrights f-value, showing that mildly deleterious genes are actually retained even in natural populations of haplo-diploid organisms.

We happened to discover that the cauliflower mosaic virus (CaMV) 35S promoter inserted into a recombinant Autographa californica multicapsid nucleopolyhedrovirus (rAcMNPV) was strongly activated during the replication of the recombinant virus in Spodoptera frugiperda (Sf9) cells. The expression of the luciferase gene from the 35S promoter in rAcMNPV was remarkably increased late in infection and was resistant to alpha-amanitin treatment. Primer extension indicated that transcriptional initiation from the 35S promoter in Sf9 cells occurred within one of the two baculoviral late promoter TAAG motifs located in the vicinity of the transcription start site in plant cells. These observations suggested that the CaMV 35S promoter served as a transcription start site for AcMNPV-induced RNA polymerase.

Lepidopteran species have a relatively high number of small holocentric chromosomes (Bombyx mori, 2n = 56). Chromosome identification has long been hampered in this group by the high number and by the absence of suitable markers like centromere position and chromosome bands. In this study, we carried out fluorescence in situ hybridization (FISH) on meiotic chromosome complements using genetically mapped B. mori bacterial artificial chromosomes (BACs) as probes. The combination of two to four either green or red fluorescence-labeled probes per chromosome allowed us to recognize unequivocally each of the 28 bivalents of the B. mori karyotype by its labeling pattern. Each chromosome was assigned one of the already established genetic linkage groups and the correct orientation in the chromosome was defined. This facilitates physical mapping of any other sequence and bears relevance for the ongoing B. mori genome projects.
