  • We are studying intracellular trafficking using mammalian cells. During my research in Japan, France, and the United States, I found that ArfGAP proteins, which were originally thought to simply inhibit membrane trafficking, can actively regulate cargo sorting and promote intracellular transport. our current research focuses on understanding how diverse exosome populations are generated and whether their formation pathways can be classified based on the functions of ArfGAP proteins.

    More recently, we have become interested in developing technologies to artificially control intracellular cargoes and organelles. By combining cell biology with materials science and physics, we aim to establish the emerging field of organelle engineering, where organelles can be manipulated using magnetic nanoparticles, external magnetic fields, and physical energies such as plasma.

    4開膜貫通タンパク質CD63はクラスリンアダプターAP-3に結合するが、エクソソームの積荷でもある。エクソソームは多胞体(MVB)が細胞膜と融合し内腔小胞(ILV)が輸送されたものと考えられる。CD63のMVBへの輸送に関与するArfGAPをスクリーニングしたところ、ADAP1とARAP1が見つかった。ADAP1とARAP1の欠損細胞ではMVBへ輸送されるEGFRの輸送には影響がなかったため、EGFRとCD63は異なるMVBに輸送され、ADAP1とARAP1はCD63の輸送にのみ関与することが明らかとなった。

    止血因子vWFはクラスリンアダプターAP-1の欠損によって分泌顆粒」WPBへの輸送が阻害されることが知られる。vWFのWPBへの輸送に関与するArfGAPをスクリーニングしたところ、SMAP1とAGFG2が見つかった。SMAP1欠損ではWPBが粒状になるが、AGFG2欠損では逆にWPBが長くなり、vWFの分泌が阻害されることがわかった。
