電界撹拌技術による迅速な酵素結合免疫吸着測定法(ELISA)の開発

Since enzyme-linked immunosorbent assay (ELISA) requires time for antigen-antibody reaction, it generally requires a long time of 1 to 12 hours or more, and the problem is that it cannot be used as rapid test.
In this study, we aim to speed up ELISA using electric field mixing (EFM). Specifically, we developed the microplate for electric field mixing with versatility in mind, examined an ELISA protocol using EFM, and investigated a new evaluation method suitable for miniaturization, replacing absorbance evaluation.
As a result, we found that rapid ELISA method is possible by using a glass plate and performing EFM at frequency that shows vigorous stirring behavior and by applying EFM not only to the antigen-antibody reaction process but also to the color development process. Also, regarding the evaluation of ELISA, we found that it is possible to evaluate even minute amounts by evaluating "saturation" using a light quality sensor instead of the conventional absorbance measurement.
