Breeding of tomato teaching materials for genetics study
Periods of research:
2018.10-NowKeywords : genetics, teaching material, tomato, Mendel
Style of Research: Individual

Tomatoes (cherry tomatoes) are selfu-polinated plants and easy to cross-polinate. They are verypopular among everyone and include well known wide range of genetic diversity like fruit size and fruit colors. We are now breeding tomato lines suitable for genetics study in elementary ~ senior high schools.

Cookinng tomato or tomato for cooking is not popular in Japan because tomatoes in Japan are bred and cultivated mainly for salad. We are trying to increase the consumption of tomato for cooking using newly released determinate-type early maturing tomato cultivar 'SUZUKOMA'.

We have found a unique late-bolting genetic resources and released a breeding material 'Tsukena Chukan Bohon Nou 2 Gou'.
Using this unique late bolting characteristics, we have analysed a mechanism of BrFLC with DNA markers. we are going to release late bolting F1 cultivars of Chinese cabbage near future.

In cool climates, Early-winter Direct-sowing cultivation is one of the important options to improve rice productivity. To help further promotion of this technique, we evaluated the impact of chemical applications to rice seeds and harvest year on the seedling establishments. The most effective chemical was Thiuram ("Kihigen-R2-Furoable";KG) among 26 combinations of chemical and material applications, which improved approximately 17 points compared to the percent seedling establishment of non-treated control seeds (3-34.9%). In addition, harvest year strongly affected the seedling establishment, and the seeds harvested in the same year as seeding was better than the seeds harvested in the previous year, where the seeds harvested in the same year showed greater dormancy than the seeds harvested in the previous year. In fact, an artificial dormancy-break treatment severely reduced the seedling establishment. Thus, we showed that "KG" improves rice-seedling establishment, and suggested the importance of seed dormancy to attain high seedling establishment in the Early-winter Direct-sowing cultivation.

As cultivated tomatoes are autogamous (or self-fertilizing) plants, we had cultivated commercial cherry tomato F1 cultivars `Orenji Kyaroru (Sakata Seed Co.), Aiko (Sakata Seed Co.) and Tyinkar Beru (Nanto Seed Co.)' then got self-pollinated F2 seeds from the harvested fruits. We have cultivated 104, 70 and 41 F2 generation individuals respectively with 10.5 cm diameter small pots. In these F2 individuals, we observed clear genetically segregated characters of leaf colors with growth degree, flesh colors, pericarp colors. Especially in the case of `Orenji Kyaroru F2', it showed three Mendel's laws, i.e. the law of dominance, segregation and independence. We conclude that commercial cherry tomato F1 cultivars are good teaching materials for genetics because you cultivate them and can easily get F2 seeds which enable to cultivate sufficient quantity of individuals to observe genetic phenomena. In scientific course of a high school, they cultivated `Orenji Kyaroru F2' and successfully observed genetic segregations. A survey by questionnaire showed that they have become more interested in genetics and understood Mendel's laws more deeply. It is an advantage to use commercial cherry tomato F1 cultivars because you don't need to cross or hybridize which is somewhat complicated procedure. Another advantage is that we can observe genetic segregation of leaf colors easily within two or three weeks after sowing. At the same time, it is a great disadvantage that we cannot use the parents of the F1 cultivars because they are never allowed to be taken out from the seed companies.

Poor seedling establishment is a major problem for Early-Winter Direct-Sowing cultivation in rice. We examined the effects of seed coating with three materials (iron, calcium peroxide, starch) on the seedling establishment. The seedling establishment rate was only 1~3% in the control without seed coating in both 2016/2017 and 2017/2018 seasons in Iwate Prefecture. Only the coating with iron showed a significant increase in the seedling establishment rate, that is, 24% in the 2016/2017 season and 11~30% in the 2017/2018 season. Significant improvement of seedling establishment by seed coating with iron was confirmed similarly in additional four Prefectures (Hokkaido, Aomori, Akita and Mie) in 2017/2018 season. We concluded that the seed coating with iron is a promising methodology for improving seedling establishment for Early-Winter Direct-Sowing cultivation in rice.

Boiled green leaves are bleached when exposed to light. To reduce this bleaching, the appearance and chemical components in 48 spinach cultivars with very deep green leaves were studied under different cultivation conditions. We observed large varietal difference in the growth rate, chlorophyll and ascorbic acid contents, and Brix.
